Clinical Study of the Efficacy of Lamotrigine^(¢ç) in Patients with Intractable Partial Seizures


Abstract
Lamotrigine(LTG) is one of newly-developed antiepileptic drugs those were developed under the basic insight of seizure mechanism. Major mechanism of LTG on the seizure inhibition is blocking of voltage-dependent sodium channel and resulting inhibition of glutamate release. After the experience of 12-weeks trial of LTG on the twenty refractory partial seizures, we found moderate benefit on seizure reduction(25.8%). Seizure reduc-tion was more marked in patients with simple partial seizures and also in patients without valproic .acid. Major side effects of LTG were associated with central nervous system -related symptoms. The frequency of these side effects were as follows; vertigo(30%), dip-lopia(20%), headache and ataxia(10% each), somnolence(5%). But all the side effects were transient and were well tolerated by the patients. The results suggested that the LTG may be useful in patients with refractory epilepsy.
